Karim Labib’s group studies the regulation and mechanisms of action of the eukaryotic replisome. Our work ranges from biochemical reconstitution of replication with purified proteins, to genetics and cell biology. Though most of our work uses the budding yeast Saccharomyces cerevisiae to address the fundamental principles of eukaryotic chromosome replication, we also use the early embryo of the nematode Caenorhabditis elegans to screen for factors that regulate the replisome in higher eukaryotes. In addition, we are now using the power of CRISPR-Cas9 genome editing in mouse embryonic stem cells to develop new models for studying the mammalian replisome.

Some of the aims of new projects could include (but are not limited to):

– identifying and characterising factors that regulate disassembly of the CMG helicase during termination of chromosome replication in higher eukaryotes.
– studying the mechanisms of termination of DNA replication.
– structural characterisation of intermediates in helicase disassembly.
– studying novel regulation of the eukaryotic replisome by post-translational modifications.

– studying the regulation of the replisome by checkpoint kinases in vivo.
